Integrated transcriptome and proteome analysis reveals potential mechanisms for differential abdominal fat deposition between divergently selected chicken lines

چکیده
Genetic selection for meat production performance of broilers concomitantly causes excessive abdominal fat deposition, accompanied by several adverse effects, such as the reduction feed conversion efficiency and reproduction performance. Our previous studies have identified important genes regulating chicken using Northeast Agricultural University broiler lines divergently selected content (NEAUHLF) an animal model. However, molecular mechanism underlying deposition differences between lean remains largely unknown. Here, we integrated transcriptome (RNA-Seq) quantitative proteome (isobaric tags relative absolute quantitation, iTRAQ) profiling analyses on tissues from NEAUHLF lines. Differentially expressed (2167 DEGs, corrected p-value < 0.01) differentially abundant proteins (199 DAPs, 0.05) were in line compared to line. Down-regulated DEGs DAPs mainly enriched pathways related fatty acid metabolism, biosynthesis, PPAR signaling, interestingly, up-regulated both lysosome pathway. Moreover, numerous key involved long-chain uptake, situ lipogenesis (fatty cholesterol synthesis), lipid droplet accumulation discovered after analysis. Excessive critically affects health economic loss producers, but is still unclear chicken. We DEGs/DAPs potential through integration analyses. findings will facilitate a better revealing provide novel insight into discrepancy

Estimating the genetic parameters for liver fat traits in broiler lines divergently selected for abdominal fat.
Intensive selection of broilers for improved growth rate is known to exert a negative effect on broiler health, such as an increase in body fat (and its related diseases). Excessive fat deposition in the liver can cause fatty liver hemorrhagic syndrome (FLHS); in addition, traits associated with liver fat have also been associated with FLHS. This study explored the genetic relationships among l...
متن کاملExpression Profiling of Preadipocyte MicroRNAs by Deep Sequencing on Chicken Lines Divergently Selected for Abdominal Fatness
Through posttranscriptional gene regulation, microRNA (miRNA) is linked to a wide variety of biological processes, including adipogenesis and lipid metabolism. Although miRNAs in mammalian adipogenesis have been worked on extensively, their study in chicken adipogenesis is still very limited. To find miRNAs potentially important for chicken preadipocyte development, we compared the preadipocyte...
متن کاملDifferential fecal microbiota are retained in broiler chicken lines divergently selected for fatness traits
Our study combined 16S rRNA-pyrosequencing and whole genome sequencing to analyze the fecal metagenomes of the divergently selected lean (LL) and fat (FL) line chickens. Significant structural differences existed in both the phylogenic and functional metagenomes between the two chicken lines. At phylum level, the FL group had significantly less Bacteroidetes. At genus level, fourteen genera of ...
متن کاملQTL analysis of body composition and metabolic traits in an intercross between chicken lines divergently selected for growth.
The high- and low-growth lines of chickens have been developed from a single founder population by divergent selection for body weight at 56 days of age for more than 40 generations. The two lines show a ninefold difference in body weight at selection age and several interesting correlated selection responses such as altered body composition and metabolic differences. We have generated a recipr...
متن کاملGenetic parameters for levels of natural antibodies in chicken lines divergently selected for specific antibody response.
The present study estimated the heritability of natural antibody (NAb) levels binding rabbit red blood cells at the day of immunization with SRBC (NAb0) and 5 d postimmunization (NAb5) in 2 chicken lines divergently selected for specific antibody levels (SpAb) against SRBC and a randombred control line. In addition, genetic correlations between the levels of NAb binding rabbit red blood cells a...
